In this study, we use Spatial light interference microscopy (SLIM), a quantitative, label-free imaging technique, to study E. coli, phages, and their interaction. The first part of the study involves imaging of infected, fluorescently-labelled E. coli cells. Bacterial genome and prophage were labelled with fluorescent proteins. The SLIM images overlay very well with the fluorescence images, proving that SLIM can detect subcellular features of E. coli. The second part of the study involves imaging isolated phages. Again, using fluorescence as ground truth, our results show that SLIM is able to detect and locate individual bacteriophages.
